Pre-adsorption of Serotonin antisera, diluted according to the antibody specification sheet, with 20 µg/ml Serotonin/BSA conjugate following the instructions below provides complete blockage of Serotonin immunolabeling. The conjugate is provided as 50 µg of lyophilized Serotonin creatinine sulfate coupled to BSA with paraformaldehyde. Also, this antiserum contains 0.09% sodium azide. Please read the instructions carefully before beginning the procedure.
The conjugate control for serotonin is intended for the immunoadsorption of serotonin antisera, catalog numbers 20079 and 20080. Preadsorption of Serotonin antisera, diluted according to the antibody specification sheet, with 20 g/ml Serotonin/BSA conjugate following the instructions below provides complete blockage of serotonin immunolabeling.
The conjugate is provided as 50 g of lyophilized serotonin creatinine sulfate coupled to BSA with paraformaldehyde.
Refer to the Instruction Manual available online at www.immunostar.com for information on tissue preparation, immunostaining techniques, troubleshooting, and formulas.
I used the 5HT-BSA conjugate to run a pre-adsorption control for the rabbit anti-serotonin antibody (#20080). 40mg/ml resulted in only very weak labeling of 1:4,000 and 1:10,000 dilutions of the rabbit anti-serotonin antibody, while all labeling was eliminated for dilutions of 1:20,000 and lower.
Dilutions of 1:4,000 to 1:80,000 all provided bright labeling in positive controls in which the BSA-5HT conjugate was omitted. The application was immunocytochemistry of sectioned brain tissue from the bumblebee Bombus impatiens (see Paulk et al 2009, J. Comp. Neurol.).
This product contains the preservative sodium azide. The concentration percent of sodium azide is ≤ .09%. Although this hazardous substance is a concentration below that required for the preparation of a Material Safety Data Sheet, we created a standard MSDS for your records.
